Small icon animations are usually of tiny small file size so there shouldn't be any major slowing down, however it may also be affected by how many requests the browser needs to make to how many servers to retrieve the graphics.
Obviously if people start adding larger graphics to their sigs then there might be cause for concern.
Regarding customised smileys, It's probably possible to replace some of the existing ones. I might experiment with this later by replacing the smileys that are least used with some Knightmare style ones to see how things go.
